Factor-VIII-related antigen: multiple molecular forms in human plasma.
Analysis of Factor-VIII-related antigen (vWAg) in normal plasma by crossed immunoelectrophoresis reveals an asymmetric pattern indicative of molecular heterogeneity. This asymmetric pattern apparently results from the presence of multiple molecular forms of VWAg with distinct, though partially overlapping, mobilities on crossed immunoelectrophoresis. Evidence for this conclusion has been obtain...
متن کاملIsolation and characterization of human factor VIII: molecular forms in commercial factor VIII concentrate, cryoprecipitate, and plasma.
Human factor VIII has been isolated from a high purity factor VIII concentrate by immunoaffinity chromatography and HPLC on Mono Q gel. Two fractions of factor VIII were obtained with a specific activity of approximately equal to 7000 units/mg. The major fraction contained eight peptide chains of 200, 180, 160, 150, 135, 130, 115, and 105 kDa plus one doublet chain of 80 kDa. The minor fraction...
متن کاملLocalization of factor-VIII-related antigen in human vascular subendothelium.
Factor-VIII-related antigen has previously been shown to be synthesized by vascular endothelial cells. Using both an immunofluorescent staining technique and electron microscopy, we have demonstrated the presence of factor-VIII-related antigen in human vascular subendothelium. This finding may have implications in the mechanism of platelet adhesion to deendothelialized blood vessel surfaces.
متن کاملApparent molecular weight of purified human factor VIII procoagulant protein compared with purified and plasma factor VIII procoagulant protein antigen.
We have compared apparent molecular weights of purified factor VIII procoagulant protein (VIII:C) and VIII:C antigen (VIII:CAg) by two different NaDodSO4 gel electrophoretic techniques. In a discontinuous NaDodSO4-7.5% polyacrylamide system, reduced and unreduced VIII:C, purified from commercial factor VIII concentrates by a monoclonal antibody immunoadsorption technique, showed a major doublet...
متن کاملElectrophoretic studies of human factor VIII-related antigen [proceedings].
I I the inherent lytic rate of cross-linked and non-cross-linked clots. To investigate this, a fibrin-plate techique was used in which small equal volumes of haemoglobin solution were spotted on to the plates instead ofthestreptokinase solution, and therateof diffusion of the haemoglobin into the plates was estimated. Yet again this was unaffected by crosslinking, and, since streptokinase has an...
